yes, vtkMassProperty should solve this problem
subtracting the mass covered by the inner surface from the mass covered by the outer surface results in the mass between the two surfaces. (as long as the surfaces do not intersect.)
btw: are we talking about cortical surfaces?

yes, pretty much, vtk can help you (was it something called mass
properties?)
