You seem to be confused between VTK and ITK.


VTK is a visualization toolkit.

ITK is an image processing toolkit with
    emphasis in segmentation and registration.
    http://www.itk.org


Segmentation in VTK can only be done by
extracting isosurfaces from the image data.
The methods iso-surface extraction are
discussed in the message below.

ITK provides a large set of segmentation
methods. Some of them are treated in the
Segmentation chapter of the SoftwareGuide.
