[ITK] 3d volume construction with itk/vtk

Hello,
I'm a new user to ITK and a beginner to medical image processing. I'm
working on a task of constructing a 3d volume from a series of micro-CT
brain slices, the volume will be used for registration in later stage. What
preparations do I make on those 2-d images? noise reduction? Do I have to
segment tissue? Thank you.
